Quercetin Affects Ceramide but not Sphingosine-1-phosphate in ras-transformed 3T3 Fibroblasts

Investigations were carried out on the effect of quercetin level major sphingolipid metabolites in ras-transformed 3T3 fibroblasts. Quercetin is a polyphenol significant biomedical importance, which has been reported to show beneficial effects such as antioxidant, anti-neoplastic, anti-ageing, etc. Sphingolipids are functionally active lipid molecules, regulate important cellular processes, like proliferation, apoptosis and transmembrane signalling, among others. Despite numerous investigations devoted influence metabolism various types cancer cells, mechanism this pathways still remains unclear. Our studies showed that sphingomyelin was decreased fibroblasts, whereas ceramide, pro-apoptotic factor, increased result elevated neutral sphingomyelinase activity. However, sphingosine-1-phosphate, related cell not altered treatment, an unexpected finding when compared our previous with cells. In addition, observed changes balancing enzyme pathway, sphingosine kinase 1, produces statistically quercetin-treated oncogene-transformed The presented data useful for better understanding flavonoids regulation could help development complex antitumour therapeutic approaches, involving natural agents polyphenols.
